Description of Research: The overall objective of this application is to fully explore the potential of auranofin as a therapeutic agent against H. pylori using a collection of antibiotic sensitive and resistant clinical isolates and to determine its capacity for individual and combination therapy as well as the potential emergence of resistance phenotypes. We also plan to further optimize auranofin activity through custom formulations. Our long-term goal is to validate auranofin as a repurposed drug for H. pylori that could be easily moved into the clinic as an alternative antibiotic as well as for other organisms that share similar vulnerabilities. This may lead to the development of a new class of drugs that can be used to treat pathogens resistant to standard antibiotic therapies
Relevance to VA: American military personnel are frequently exposed to H. pylori at sites that have higher rates of infections, acquiring virulent strains that are harder to treat, which are also associated with carcinogenesis. In a recent study conducted among US Veterans the prevalence of active H. pylori remains high (28.9%) with even higher rates in Veterans of African American or Hispanic descent. Because of increased antibiotic resistance, particularly the emergence of clarithromycin and metronidazole resistance strains, new treatments are needed.
